Accuracy & Precision - Part 3 - Role of advance features to minimize carry-over &/or cross-over in fully auto clinical chemistry analyzer
As we have discussed in part 2 of this blog, “Advanced Washing Protocol” & “Type of Probe / Mixer” played an important role to minimize carry-over &/or cross-over in fully auto clinical chemistry analyzer.
Majority of fully auto clinical chemistry analyzers are equipped with daily or routine wash protocol to wash cuvettes with “DI-Water” as well as “Detergent Wash”. Even air purge with fresh water is done by majority of analyzers to remove air bubble from dispensing tubes. Cuvette washing station with 2x4 or 2x6 steps automatic washing with automatic cleaning and drying of reaction cuvettes is also important to reduce carry-over &/or cross-over.
In this blog will talk about couple of advance and unique features available in select few analyzers only, which also plays important role to minimize carry-over &/or cross-over, as mentioned below. . .
- Carry-over Evasion Program - This is a programmed pre-wash &/or post-wash protocol to avoid carry-over of sample &/or reagent specifically to avoid interference. For example, Icteric samples (yellow pigment of bilirubin) can absorb light spectrally as well as chemically interfere with peroxidase of other test reagent. Same way, some reagents are having a tendency to react with each other. i.e. substrate of one reagent can react with the substrate of other reagent. Carry-over evasion program facilitate extra washing with detergent as well as with the reagent of 2nd test (which may get influence from interfering sample or reagent).
- This extra wash with detergent &/or reagent between two tests of interfering samples / reagents will reduce the chances of cross reactions. Carry-over evasion program will do extra washing for probe as well as reaction cuvette.
- Test Order Selection - These feature allows selection of test order in such a way to avoid tests of interfering substances after each other. i.e. Icteric samples may react with the reagent having peroxidase, in this situation carry-over evasion program will not perform test having peroxidase, immediately after Icteric sample. So, this program allows you to set the test order to prevent cross reaction of different reagents having tendency to react with each other.
